演讲题目:Study of RNA virus-host interaction by multi-omics approaches

演讲大纲:
Infection of emerging or re-emerging RNA viruses, such as influenza A virus and enteroviruses, could result in severe diseases in humans. In the infected cells, the biological processes are altered by either the event of infection or viral protein expression. Influenza A virus is a respiratory virus that could cause pandemics in history. The NS1 protein of influenza A virus antagonizes cellular antiviral response through various mechanisms. To globally study the cellular functions that are affected by the NS1 protein, we applied a proteomic approach to identify host proteins that may associate with the NS1 protein. The results showed that several proteins involved in RNA processing and protein targeting, respectively, could interact with the NS1 protein. In addition to influenza A virus, human enterovirus type A71 (EV-A71) could cause severe neurological complications in children. Therefore, we used a similar approach to investigate the biological impacts of the RNA-dependent RNA polymerase (RdRP, also denoted as 3D protein) encoded by EV-A71 on host cells. Moreover, since viral infection can dysregulate gene expression in host cells, we applied the single-cell RNA sequencing on EV-A71-infected cells to analyze the gene dysregulation at a single-cell level. In summary, we have used proteomics and transcriptomic approaches to study the virus-host interaction that may be impacted on viral replication and pathogenicity.
